Add 1.28g (0.0072mol) of 4-(pyridin-2-yl)thiazol-2-amine into a 50mL three-necked flask, and dissolve 10ml of tetrahydrofuran. Then use 10 mL of dry tetrahydrofuran to dissolve 4-methyl-1,2,3-thiadiazole-5-formyl chloride, and slowly add it dropwise at a temperature lower than -5°C. After the addition is complete, add 5 mL of triethylamine and stir at room temperature. After reacting for 12 hours, the solid was filtered off, and the solvent was evaporated under reduced pressure to obtain a yellow oily liquid.

Add a small amount of DCM to the oily liquid and shake while adding until a large amount of solid precipitates. Filter out the solid, and wash the filter cake with 10% sodium hydroxide aqueous solution, water, and saturated sodium chloride aqueous solution in sequence. Dry over anhydrous sodium sulfate to obtain the title compound as a yellow solid 0.99g, yield 41.1%, melting point 179-180°C. 1HNMR (MHz, DMSO-d6): δ13.40 (brs, 1H), 8.63 (m, 1H), 8.01 (m, 3H), 7.37 (m, 1H), 2.89 (s, 3H). HRMS(ESI)C13H10ON4ClS2[M+H]+Theoretical value is 336.99791, and the experimental value is 336.99807.

Application and preparation of 4-(pyridin-2-yl)thiazol-2-amine

The preparation of 4-(pyridin-2-yl)thiazol-2-amine is as follows: add 2.0g (0.01mol) of α-bromo-2-pyridine ethanone and 0.76g of thiourea ( 0.01mol) and 50mL ethanol, reflux reaction for 4h. After the reaction is completed, the ethanol solvent is evaporated under reduced pressure. Column chromatography separation, using DCM:MeOH:NH4OH (20:1:0.1, v/v=50:1:0.1) as the mobile phase. The title compound 4-(pyridin-2-yl)thiazol-2-amine was obtained as a white solid 1.41 g, yield 79.6%, melting point 178-179°C. 1HNMR (MHz, DMSO-d6): δ8.54(m, 1H), 7.82(m, 2H), 7.25(m, 2H), 7.11(brs, 2H).MS(ESI)m/z[M+H ]+199.97.
