Background and overview of preparation and application of (5-hydroxy-3-pyridyl)boronic acid

(5-Hydroxy-3-pyridyl)boronic acid is a boronic acid derivative. Boronic acid derivatives are widely used in organic synthesis for the formation of bromopyridine for carbon-carbon bonds. In the Suzuki coupling, aryl halides and aryl or vinyl borates or boronic acids are coupled using Pd(PPh3)4. There are reports in the literature that it can be prepared from 3-bromo-5-hydroxypyridine as raw material through the traditional borate ester synthesis method.

Preparation and application of (5-hydroxy-3-pyridyl)boronic acid

To a solution of 3-bromo-5-hydroxypyridine (1eq, 3.56mmol) in MeCN (10ml), pinacol diborate (1.2eq, 4.27mmol, 1.11g) and KOAc (3eq, 10.7mmol, 1.05g) and Pd(dppf)2Cl2 (0.04eq, 0.142mmol, 105mg). The mixture was heated with microwave irradiation at 160°C for 15 minutes. The mixture was filtered to obtain the title compound

(5-hydroxy-3-pyridyl)boronic acid can be used to prepare compounds having the following pyrazolo[3,4-d]pyrimidine structure. Pyrazolo[3,4-d]pyrimidine compounds are nitrogen-containing fused ring compounds, which are isosteres of purine, and show a variety of pharmacological effects because they have a similar structure to purine. Therefore, pyrazolo[3,4-d]pyrimidine compounds have always been a research hotspot in the field of pharmacology. It has been reported that pyrazolo[3,4-d]pyrimidine compounds have various pharmacological activities such as antibacterial, antiviral, anticancer, anti-inflammatory, antihypertensive, antidiabetic, inhibiting xanthine oxidase and thereby treating gout.
